The usual suspects with the glo plug system is individual plugs (one in each cylinder) beginning to increase resistance or burning out - open circuit. There is a glo plug control module which can also fail. It is relatively easy to check individual glo plugs with an ohmmeter but you should expect very low resistance - on the order of 0.1 to 0.2
On both the 4 cylinder and the V6 engines the glow plugs are controlled by J179 - Automatic glow period control unit which is connected to J623 - Engine control unit. If by " comfort control module" you are referring to J393 - Convenience system central control unit then the answer is no, the car will no longer start.

The only wrinkle - for those not used to Audi connectors - is that it was easier for me to remove the connected fuel pump module first, and then disconnect the connector from the module after it was out. Otherwise, the connection tab is on the underside of the module, and the module is screwed into some sheet metal, so it was difficult to get to.
